Adult Heart Mounting

Adult heart mounting is a biological preparation technique that secures an excised adult heart in a defined orientation for dissection, imaging, or downstream analysis. The procedure supports the tissue while preserving its anatomical relationships, allowing researchers to expose chambers, valves, vessels, or myocardial surfaces without excessive deformation. Proper mounting improves consistency in microscopy, histological sectioning, immunostaining, and structural assessment. In cardiovascular biology, this preparation helps connect cardiac anatomy with tissue organization, disease-related remodeling, and experimental measurements, providing a reliable foundation for studying adult heart structure and function.

Use of zebrafish for cardiovascular research is expanding towards research on adult hearts. For these applications, quick and simple isolation of cardiac tissues is key to avoid post-mortem changes and to obtain an adequate number of samples. Here, we describe a fast and reproducible method for dissecting adult zebrafish hearts.

We present a method to isolate the adult organ of Corti as three intact cochlear turns (apex, middle, and base). We also demonstrate a procedure for immunostaining with fluorescently tagged antibodies. Together these techniques allow the study of hair cells, supporting cells, and other cell types found in the cochlea.
