Angiotensinogen Precursor Protein

Angiotensinogen precursor protein is a liver-produced, circulating glycoprotein that supplies the starting material for the renin-angiotensin-aldosterone system, a hormonal pathway that regulates blood pressure and fluid balance. When kidney blood flow falls or related signals activate renin release, renin cleaves angiotensinogen to form angiotensin I; angiotensin-converting enzyme then generates angiotensin II, which promotes vasoconstriction and aldosterone secretion. Studying angiotensinogen expression, processing, and genetic variation helps explain cardiovascular and renal physiology, investigate hypertension, and evaluate therapies that interrupt this pathway. Its precursor role makes it an important subject in molecular biology and disease research.

Newly synthesized precursors are inhibited from spontaneously folding into their native conformation by the cytosolic chaperones, heat shock proteins 70 (Hsp70), and mitochondrial import stimulation factors (MSFs). Purification and Aggregation of the Amyloid Precursor Protein Intracellular Domain

A method for large-scale purification of the APP intracellular domain (AICD) is described. We also describe methodology to induce in vitro AICD aggregation and visualization by atomic force microscopy. The methods described are useful for biochemical/structural characterization of the AICD and the effects of molecular chaperones on its aggregation.

Differentiation of Embryonic Stem Cells into Oligodendrocyte Precursors

We describe a small molecule-based protocol for differentiation of mouse embryonic stem cells into oligodendrocyte precursor cells (OPCs). This protocol generates Olig2+NG2+ OPCs with high efficiency by 30 days of differentiation. We also describe a method to generate "spiking" OPCs that can fire action potentials.
