Arterial Sheet Culture

Arterial sheet culture is an ex vivo biology technique that maintains thin, sheet-like sections of arterial tissue under controlled laboratory conditions to study vascular structure and function. The tissue is placed in nutrient media with regulated temperature, gas exchange, and sterility, allowing cells to remain within their native cell–cell and cell–matrix organization while researchers monitor survival, migration, remodeling, or injury responses. This approach preserves interactions that isolated-cell systems may lose and supports investigations of endothelial and smooth muscle behavior, vascular repair, inflammation, and responses to experimental treatments. Arterial sheet culture therefore provides a useful bridge between simplified cell culture and more complex in vivo models.

This protocol describes a new technique that completely inverts blood vessels of interest, exposing only the endothelial side to enzymatic digestion.
