Barrier Integrity Assay

Barrier integrity assays are experimental methods used to evaluate how effectively a cell layer or tissue barrier limits the movement of substances between compartments, making them important for studying epithelial and endothelial function. They generally measure permeability to a tracer or electrical resistance across cultured cells; tight junctions and other barrier structures maintain low passage, while injury, inflammation, or altered signaling can increase permeability or reduce resistance. These assays support research on vascular and tissue barriers, disease mechanisms, drug transport, toxicity, and barrier repair, while providing quantitative readouts for comparing treatments and cell models.

Assay for Blood-brain Barrier Integrity in Drosophila melanogaster

Cited by 2 •

2019

Blood-brain barrier integrity is critical for nervous system function. In Drosophila melanogaster, the blood-brain barrier is formed by glial cells during late embryogenesis. This protocol describes methods to assay for blood-brain barrier formation and maintenance in D. melanogaster embryos and third instar larvae.

Oral Gavage in Neonatal Mouse Pups and Functional Assessment of Gut Barrier Integrity Using Ussing Chambers

This article describes: (1) oral gavage delivery of test molecules to neonatal mice as early as DOL 6; and (2) ex vivo evaluation of colonic epithelial barrier function in biopsies from DOL 10 to weaning (DOL 20) pups by assessing paracellular and transcellular permeability with Ussing chambers.

Assessing Blood-Brain Barrier Integrity in a Laser-Induced Brain Injury Rat Model

Source: Kuts, R. et al. Laser-Induced Brain Injury in the Motor Cortex of Rats. J. Vis. Exp. (2020)This video demonstrates assessing blood-brain barrier (BBB) integrity following laser-induced brain injury in rats. A tracking dye is injected into the tail vein, allowing it to circulate to the brain and leak into the tissue through the damaged BBB. The brain is then extracted and processed to release the dye. The fluorescence intensity of the dye is measured to quantify the extent of...

Maintaining blood-brain barrier coverage is key for the homeostasis of the central nervous system. This protocol describes in vitro techniques to delineate the fundamental and pathological processes that modulate blood-brain barrier coverage.

Assessing Transmissible Spongiform Encephalopathy Species Barriers with an In Vitro Prion Protein Conversion Assay
