Bisulfite Treatment Protocol

The bisulfite treatment protocol is a molecular biology method used to distinguish methylated from unmethylated cytosines in DNA, enabling analysis of DNA methylation patterns. During treatment, DNA is denatured and exposed to sodium bisulfite, which converts unmethylated cytosine residues to uracil while leaving methylated cytosines largely unchanged; subsequent amplification and sequencing preserve these differences as detectable sequence changes. Researchers use the protocol for methylation-specific PCR, bisulfite sequencing, and epigenetic profiling of genes and genomic regions. These data help investigate gene regulation, development, disease-associated methylation, and other biological processes influenced by epigenetic modification.

Bisulfite mutagenesis is the gold standard for analyzing DNA methylation. Our modified protocol allows for DNA methylation analysis at the single-cell level and was specifically designed for individual oocytes. It can also be used for cleavage-stage embryos.

Here, we present a protocol to remove aldehydes and reactive ketones from mixtures by a liquid-liquid extraction protocol directly with saturated sodium bisulfite in a miscible solvent. This combined protocol is rapid and facile to perform. The aldehyde or ketone can be re-isolated by the basification of the aqueous layer.
