Bleach Wash Protocol

A Bleach Wash Protocol is a biological preparation method that uses dilute sodium hypochlorite to remove external organisms and selectively isolate resistant structures, such as nematode eggs, from mixed cultures. During the wash, hypochlorite oxidizes and breaks down exposed organic material, while a protective egg shell can allow eggs to remain intact; repeated rinsing then removes residual bleach and dissolved debris. In Caenorhabditis elegans research, this procedure helps reduce microbial contamination and synchronize populations by recovering eggs that hatch under controlled conditions. Consistent timing, concentration, and thorough washing are essential for preserving sample viability and producing reproducible developmental experiments.

Sequential Photo-bleaching to Delineate Single Schwann Cells at the Neuromuscular Junction

Visualizing individual cells in densely packed tissues, such as terminal Schwann cells (SCs) at neuromuscular junctions (NMJs), is challenging. "Sequential photo-bleaching" allows delineating single terminal SCs, for instance in the triangularis sterni muscle explant, a convenient nerve-muscle preparation, where sequential bleaching can be combined with time-lapse imaging and post-hoc immunostainings.

After filtration, the precipitate is washed to remove coprecipitated impurities and any remaining mother liquor.
