Blood Preparation

Blood preparation is the controlled collection and processing of blood samples to preserve and separate their cellular and liquid components for biological analysis. Depending on the intended test, blood is collected with or without anticoagulants, then handled through steps such as mixing, centrifugation, plasma or serum separation, and preparation of blood smears. These procedures help maintain cell morphology and prevent clotting, contamination, or other changes that could distort results. Properly prepared blood supports hematology, clinical diagnostics, immunology, molecular biology, and biomedical research by providing reliable material for examining blood cells, proteins, pathogens, and other indicators of health and disease.

En Face Preparation of Mouse Blood Vessels

A procedure for making en face preparations of the mouse carotid artery and aorta is described. Such preparations, when immunofluorescently stained with specific antibodies, enable us to study localization of proteins and identification of cell types within the entire vascular wall by confocal microscopy.

Establishment of human models of the blood-brain barrier (BBB) can benefit research into brain conditions associated with BBB failure. We describe here an improved technique for preparation of a contact BBB model, which permits coculturing of human astrocytes and brain endothelial cells on the opposite sides of a porous membrane.

The preparing and processing of dried blood spots (DBS) for their final analysis are still poorly standardized for most diagnostic applications. To overcome this shortcoming, a comprehensive step-by-step protocol is suggested and subsequently evaluated with regard to its effectiveness for detecting markers of viral infections.
