Blue Native Page

Blue Native Page is a non-denaturing gel electrophoresis technique that separates intact protein complexes while preserving many of their native interactions and activities. In this method, Coomassie Brilliant Blue binds proteins and provides a negative charge, allowing complexes to migrate through a polyacrylamide gel according to their size, shape, and charge rather than being fully dissociated by detergents such as SDS. In biology, Blue Native Page helps analyze membrane protein assemblies, respiratory-chain complexes, oligomeric states, and supercomplex formation. Researchers can combine it with in-gel activity assays or second-dimension SDS-PAGE to assess complex composition and function.

In this video, we describe the characterization of multiprotein complexes (MPCs) by blue native polyacrylamide gel electrophoresis (BN-PAGE). In a first dimension, dialyzed cellular lysates are separated by BN-PAGE to identify individual MPCs. In a second dimension SDS-PAGE, MPCs of interest are further subdivided to analyze their constituents by immunoblotting.

Resolving Affinity Purified Protein Complexes by Blue Native PAGE and Protein Correlation Profiling

Here we present protocols for affinity purification of protein complexes and their separation by blue native PAGE, followed by protein correlation profiling using label free quantitative mass spectrometry. This method is useful to resolve interactomes into distinct protein complexes.

In this video, we demonstrate an electrophoretic technique, called blue native PAGE, for the analysis of mitochondrial respiratory protein complexes in their natively folded and functionally active state. The anionic Coomassie blue dye causes the protein complexes to become negatively charged, allowing separation based on size and structure.

A protocol for the elucidation of plant thylakoid protein complex organization and composition with blue native polyacrylamide gel electrophoresis (BN-PAGE) and 2D-SDS-PAGE is described. The protocol is optimized for Arabidopsis thaliana, but can be used for other plant species with minor modifications.
