Calcein Alizarin Red Labeling

Calcein Alizarin Red labeling is a fluorescent double-labeling technique used to visualize and measure new mineral deposition in bone and other calcifying tissues. Calcein and Alizarin Red bind calcium and become incorporated at mineralization fronts; when administered at separate time points, they produce distinct green and red bands that mark the spatial progression of mineral formation. Microscopy of the labeled tissue reveals the distance between bands, allowing researchers to calculate mineral apposition and bone formation rates. The method supports studies of skeletal development, bone remodeling, osteoporosis, fracture healing, osteogenic cell activity, and the performance of biomaterials or regenerative therapies.

The cultured osteoblasts are first stained and then the dye is recovered for colorimetric quantification of calcium deposits or the degree of osteoblast mineralization.

Here, we have used alizarin red staining to show that lead acetate exposure causes a bone mass change in zebrafish larvae. This staining method can be adapted to the investigation of bone loss in zebrafish larvae loss induced by other hazardous toxicants.
