Cell Division Tracking

Cell division tracking is the observation and recording of when, where, and how cells progress through division, enabling researchers to follow cellular lineages over time. In biology, it commonly combines fluorescent labels or genetically encoded reporters with time-lapse microscopy to identify mitotic events, measure division timing, and trace daughter cells across successive generations. These measurements reveal how cell-cycle regulation, tissue organization, and developmental signals influence proliferation. Cell division tracking also supports studies of stem cells, cancer biology, tissue regeneration, and drug responses by linking individual cellular behaviors to population-level growth and fate.

Optimized Staining and Proliferation Modeling Methods for Cell Division Monitoring using Cell Tracking Dyes

Successful use of cell tracking dyes to monitor immune cell function and proliferation involves several critical steps. We describe methods for: 1) obtaining bright, uniform, reproducible label-ing with membrane dyes; 2) selecting fluorochromes and data acquisition conditions; and 3) choosing a model to quantify cell proliferation based on dye dilution.

Preparation of Onion Root Tips and Solutions ExpandFor the cell cycle experiment using root tips, first, leave an onion suspended over a beaker of water to grow roots for several days. Then, clean the onion roots of any dirt or debris. Next, in a 1.5 mL tube, dissolve 10 mcg of nocodazole per 1 mL of dimethyl sulfoxide solution, making 1 tube per student.
