Cell Membrane Structure

Cell membrane structure refers to the organization of lipids, proteins, and carbohydrates that forms the boundary surrounding every cell, maintaining internal conditions while enabling communication with the environment. A phospholipid bilayer creates a flexible, selectively permeable barrier: hydrophobic tails face inward, while hydrophilic heads interact with water, and membrane proteins regulate transport, signaling, adhesion, and energy conversion. Carbohydrate groups attached to lipids and proteins help cells recognize one another and interact with extracellular materials. Understanding cell membrane structure is essential for explaining diffusion, osmosis, receptor activity, cellular signaling, pathogen entry, and the action of drugs that target membrane components.

β-barrel outer membrane proteins (OMPs) serve many functions within the outer membranes of Gram-negative bacteria, mitochondria, and chloroplasts. Here, we hope to alleviate a known bottleneck in structural studies by presenting protocols for the production of β-barrel OMPs in sufficient quantities for structure determination by X-ray crystallography or NMR spectroscopy.

Procedures for complete reconstitution of a prototype voltage-gated potassium channel into lipid membranes are described. The reconstituted channels are suitable for biochemical assays, electrical recordings, ligand screening and electron crystallographic studies. These methods may have general applications to the structural and functional studies of other membrane proteins.
