Chemoattractant Gradient

A chemoattractant gradient is a spatial change in the concentration of a chemical signal that guides the directed movement of cells or organisms toward higher concentrations. Cells detect differences across their surface through chemoattractant receptors, which activate intracellular signaling that reorganizes the cytoskeleton and biases movement toward the signal source. In biology, these gradients help immune cells reach sites of infection, guide developing cells to appropriate locations, and coordinate tissue repair. Studying gradient formation, sensing, and cellular migration clarifies how local chemical cues produce organized behavior and can inform research on inflammation, development, and cancer progression.

3D Analysis of Multi-cellular Responses to Chemoattractant Gradients

We describe a method to construct devices for 3D culture and experimentation with cells and multicellular organoids. This device allows analysis of cellular responses to soluble signals in 3D microenvironments with defined chemoattractant gradients. Organoids are better than single cells at detection of weak noisy inputs.

Using frog sperm we describe and compare two classes of assays used to detect sperm chemoattraction – sperm accumulation assays and sperm tracking assays.
