Crispr-dcas9 Complex

The CRISPR-dCas9 complex is a programmable DNA-binding system that uses a catalytically inactive Cas9 protein to target specific genomic sequences without cutting the DNA. A single-guide RNA directs dCas9 to a complementary DNA region, where the complex binds through RNA-DNA base pairing and can block transcription or recruit regulatory effector proteins. In biology, this platform enables targeted gene repression or activation, epigenetic regulation, and visualization of genomic loci. Because it alters gene expression without creating double-strand breaks, CRISPR-dCas9 supports functional genomics, cellular pathway analysis, and the development of more controlled genome-engineering strategies.

CRISPR/Cas systems mediate adaptive immunity in Bacteria and Archaea. Many Cas proteins are proposed to act as endoribonucleases acting on crRNA precursors of varying length. Here we illustrate three different approaches to generate pre-crRNA substrates for the biochemical analysis of Cas endonuclease activity.
