Crypt Dynamics

Crypt dynamics describes how epithelial cells within tissue crypts are generated, maintained, displaced, and lost over time, making it central to understanding tissue renewal. In intestinal crypts, stem and progenitor cells divide near the crypt base, produce differentiating descendants, and support upward cell migration toward the villus, where cells perform specialized functions before being shed. Studying these coordinated processes reveals how stem-cell niches preserve epithelial homeostasis, respond to injury, and regulate regeneration. Crypt dynamics also provides a framework for investigating abnormalities in proliferation and differentiation associated with inflammation, aging, and colorectal cancer.

Generation of Murine Primary Colon Epithelial Monolayers from Intestinal Crypts

In this protocol, we describe how to generate murine primary epithelial colon monolayers directly from intestinal crypts. We provide experimental approaches to generate confluent monolayers on permeable filters, confluent monolayers for scratch wound healing and biochemical studies, and sparse and confluent monolayers for immunofluorescence analysis.
