Dermal Cell Isolation

Dermal cell isolation is a laboratory technique used to obtain viable cells from the dermis, the connective-tissue layer beneath the epidermis, for controlled study and culture. Typically, researchers mince dermal tissue and use enzymatic digestion to loosen the extracellular matrix, then separate released cells through filtration or centrifugation before maintaining them in culture conditions. Isolated dermal cells, including fibroblast-rich populations, support investigations of skin structure, extracellular-matrix production, wound healing, and tissue engineering. The method also provides a foundation for studying skin disorders and testing how cells respond to biomaterials, growth factors, or other experimental treatments.

Fibroblast behavior underlies a spectrum of clinical entities, but they remain poorly characterized, largely due to their inherent heterogeneity. Traditional fibroblast research relies upon in vitro manipulation, masking in vivo fibroblast behavior. We describe a FACS-based protocol for the isolation of mouse skin fibroblasts that does not require cell culture.

Cell reprogramming requires the introduction of key genes, which regulate and maintain the pluripotent cell state.
