Dermal Infection

Dermal infection is the invasion and multiplication of microorganisms within the skin, a condition that matters because it can disrupt the body’s protective barrier and cause localized or systemic disease. Bacteria, fungi, viruses, or parasites may enter through wounds, hair follicles, or other barrier defects, where immune recognition triggers inflammation, cytokine release, and recruitment of defensive cells. Studying dermal infections helps biologists understand host–microbe interactions, tissue repair, and factors that influence disease severity. This knowledge supports accurate diagnosis, wound management, antimicrobial treatment, infection prevention, and the development of therapies for drug-resistant or recurrent skin infections.

Fibroblast behavior underlies a spectrum of clinical entities, but they remain poorly characterized, largely due to their inherent heterogeneity. Traditional fibroblast research relies upon in vitro manipulation, masking in vivo fibroblast behavior. We describe a FACS-based protocol for the isolation of mouse skin fibroblasts that does not require cell culture.

Here we describe a protocol that allows histological and molecular analysis of skin samples after Candida albicans intradermal injection. This protocol maintains the structural integrity of the skin and allows for the localization of tissue-resident or newly recruited immune cells as well as the pathogen distribution.

Humanized Mouse Model to Study Bacterial Infections Targeting the Microvasculature

Neisseria meningitidis is a human specific pathogen that infects blood vessels. In this protocol human microvessels are introduced into a mouse by grafting human skin onto immunocompromised mice. Bacteria adhere extensively to the human vessels, leading to vascular damage and development of the purpuric rash typically observed in human cases.
