Dna Labeling

DNA labeling is the process of attaching a detectable tag to DNA so researchers can locate, identify, quantify, or follow specific molecules. Labels may be incorporated enzymatically as modified nucleotides during DNA synthesis or attached chemically to DNA strands, with fluorescent dyes commonly producing signals that can be measured by microscopy or other detection systems. In biology, labeled DNA supports probe-based hybridization, chromosome mapping, gene expression studies, and detection of particular sequences in research and diagnostic assays. These methods help reveal genome organization, track nucleic acid behavior, and distinguish target sequences within complex biological samples.

This protocol describes the characterization of DNA polymerase synthesis of modified DNA through observation of changes to near-infrared fluorescently labeled DNA using gel electrophoresis and gel imaging. Acrylamide gels are used for high resolution imaging of the separation of short nucleic acids, which migrate at different rates depending on size.

A method for fluorescent staining of fixed biological material with the specific DNA label methyl green is described. Methyl green is used in a diluted aqueous solution and is very resistant to photobleaching. Its far-red emission allows for deep specimen imaging, making it particularly adequate for whole embryos.

Drosophila oogenesis continues to be exceptionally useful in the study of mitochondrial proliferation and inheritance. This manuscript describes a detailed protocol used to label the replicating mitochondrial DNA (mtDNA) in Drosophila adult ovaries with 5-ethynyl-2´-deoxyuridine (EdU), which facilitates uncovering mechanisms associated with mitochondrial inheritance that were previously debatable.
