Dna Reporter Construct

A DNA reporter construct is an engineered DNA molecule that links a regulatory sequence or gene of interest to a reporter gene, enabling biological activity to be measured through a detectable signal. When the regulatory element is activated, cellular machinery transcribes and translates the reporter, producing an output such as fluorescence, luminescence, or an enzymatic color change that reflects gene expression or pathway activity. In biology, these constructs help researchers examine promoter function, transcriptional regulation, cellular responses, and gene delivery, providing quantitative or visual evidence of molecular events in living cells and experimental systems.

MicroRNAs (miRNAs) are important regulators of gene expression and have been shown to play a role in numerous biological processes. To better understand miRNA-UTR interactions, we have created a genome-wide collection of 3 UTR luciferase reporters paired with a novel luciferase gene and assay reagent, the LightSwitch system.

This article presents a detailed protocol for T4 ligation and denaturing PAGE purification of small circular DNA molecules, annealing and native PAGE analysis of circular tiles, assembling and AFM imaging of 1D and 2D DNA nanostructures, as well as agarose gel electrophoresis and centrifugation purification of finite DNA nanostructures.

Using DNA assembly, multiple CRISPR vectors can be constructed in parallel in a single cloning reaction, making the construction of large numbers of CRISPR vectors a simple task. Tomato hairy roots are an excellent model system to validate CRISPR vectors and generate mutant materials.

The genetic reporter assay is a well-established and powerful tool for dissecting the relationship between DNA sequences and their gene regulatory activities. Coupling candidate regulatory elements to reporter genes that carry identifying sequence tags enables massive parallelization of these assays.
