Acrylamide Gel Electrophoresis

Acrylamide gel electrophoresis is a laboratory technique that separates proteins or other charged biomolecules by their movement through a porous polyacrylamide matrix in an electric field. The gel acts as a molecular sieve: smaller molecules migrate more readily through its cross-linked network, while charge, molecular shape, and, in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), detergent-coated protein size determine migration. Researchers use the method to estimate molecular mass, assess sample purity, compare protein expression, and resolve components before downstream analyses such as immunoblotting or mass spectrometry. Its high resolving power makes it a foundational method in molecular and cell biology.
