Dye Loading

Dye loading is the process of introducing fluorescent or chromogenic dyes into cells, tissues, or other biological samples so their structure, activity, or chemical environment can be measured. Depending on the dye and specimen, loading occurs through passive diffusion, membrane-permeant chemical modifications, microinjection, electroporation, or carrier-mediated delivery; intracellular enzymes may then convert a probe into its active form. In biology, loaded dyes support fluorescence microscopy, cell tracking, viability assays, calcium and membrane-potential measurements, and analysis of organelle function. Careful control of dye concentration, exposure time, and toxicity improves signal quality while preserving normal cellular behavior.

Targeted-esterase induced dye loading (TED) supports the analysis of intracellular calcium store dynamics by fluorescence imaging. The method bases on targeting of a recombinant Carboxylesterase to the endoplasmic reticulum (ER), where it improves the local unmasking of synthetic low-affinity Ca2+ indicator dyes in the ER lumen.

Microelectrode-Mediated FM Dye Loading and Unloading in Drosophila Larvae Nerve Cells

In this video, Drosophila larvae nerve fibers are immersed in FM dye, which binds to cell membrane lipids. Suction microelectrode and electrical pulses stimulate the endocytosis process, incorporating the FM dye into newly formed endocytic vesicles. This method, which visualizes presynaptic stages of neurotransmission, is useful for studying synaptic function and neurotransmitter release.

Loading a Calcium Dye into Frog Nerve Endings Through the Nerve Stump: Calcium Transient Registration in the Frog Neuromuscular Junction

Here, we describe a method for loading a calcium-sensitive dye through the frog nerve stump into the nerve endings. We also present a protocol for the recording and analysis of fast calcium transients in the peripheral nerve endings.
