Embryo Fixation

Embryo fixation is the process of preserving an embryo’s structure and molecular components by treating it with a chemical fixative, allowing researchers to examine development after growth has stopped. Fixatives stabilize cells and tissues by crosslinking or precipitating proteins, limiting enzymatic breakdown and maintaining spatial relationships for microscopy and downstream staining. In biology, fixed embryos support analysis of cell division, tissue patterning, gene or protein localization, and anatomical defects through methods such as immunostaining, in situ hybridization, and fluorescence imaging. Careful control of fixation time and conditions is essential because over-fixation can reduce probe or antibody access and alter detectable signals.

Two basic types of preparation are used to visualize specimens with a light microscope: wet mounts and fixed specimens. The simplest type of preparation is the wet mount, in which the specimen is placed in a drop of liquid on the slide. A liquid specimen can be directly deposited on the slide using a dropper. Fluorescence in-situ hybridization (FISH) is often required in combination with histopathology and molecular diagnostics for selection of therapy in personalized medicine. A novel non-cross-linking, formalin-free tissue fixative that allows high quality morphologic, molecular and FISH analyses from the same specimen by addition of a post-fixation step before FISH is presented.
