Endothelial Cell Sprouting

Endothelial cell sprouting is a form of angiogenesis in which new blood-vessel branches grow from pre-existing vessels, supporting tissue development, repair, and disease progression. In response to signals such as vascular endothelial growth factor, endothelial cells loosen junctions, degrade the surrounding extracellular matrix, and migrate toward the stimulus; leading tip cells guide movement while trailing stalk cells proliferate and form the nascent vessel lumen. Researchers study this process using three-dimensional culture and ex vivo tissue assays to measure vessel formation and remodeling. These models help clarify vascular biology and evaluate therapies for cancer, ischemic disease, inflammation, and tissue engineering.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.
