Retroviral Infection

Retroviral infection is the process by which retroviruses enter host cells, convert their RNA genomes into DNA, and use cellular machinery to produce new viral particles. After entry, the viral enzyme reverse transcriptase synthesizes DNA from the RNA template, and integrase inserts this viral DNA into the host chromosome as a provirus; host transcription then generates viral RNA and proteins, which assemble and bud from the cell membrane. In biology, studying retroviral infection reveals how genetic information moves between viruses and cells, how persistent infections develop, and how viral replication can alter host-cell function. These mechanisms also support research on antiviral drugs, vaccine design, and retroviral vectors for gene delivery.

Recombinant Retroviral Production and Infection of B Cells

An efficient system of structure and function analysis of a gene in an ex vivo culture of splenic B-lymphocytes is described. This method takes advantage of recombinant retroviral production in a helper free, ecotrophic packaging cell line. Stable, heritable expression of a gene of interest within primary lymphocytes is achieved leading to generation of surface antibodies on B cells undergoing class switch recombination.
