Erv1 Oxidase

Erv1 oxidase is a flavin adenine dinucleotide-dependent sulfhydryl oxidase that catalyzes disulfide bond formation and helps maintain the oxidative environment of the mitochondrial intermembrane space. It accepts electrons from reduced cysteine residues in substrate proteins, transfers them through its FAD cofactor, and ultimately passes them to molecular oxygen or cytochrome c, while cooperating with the Mia40/CHCHD4 import pathway. This system supports the oxidative folding and mitochondrial import of intermembrane-space proteins, including proteins with twin Cx9C or related cysteine motifs. Understanding Erv1 oxidase clarifies mitochondrial protein biogenesis and cellular redox regulation.

NADPH oxidase is the major source of reactive oxygen species (ROS) in phagocytes. Because of the ephemeral nature of ROS, it is difficult to measure and monitor ROS levels in living animals. A minimally invasive method for serial quantification of ROS in living mice is described.
