Fly Housing Vials

Fly housing vials are small laboratory containers used to maintain, observe, and handle live flies under controlled conditions, supporting reproducible biological research. Their design confines insects while providing access to a food or culture medium and, when appropriate, ventilation through a secure closure, creating a manageable environment for survival and reproduction. Researchers use these vials to establish fly stocks, collect offspring, and study genetics, development, behavior, physiology, and disease-related traits. Standardized housing helps reduce environmental variation, enables careful monitoring across generations, and supports experiments involving controlled crosses, treatments, and phenotypic analysis.

This protocol describes a microinjection methodology that we have standardized and used for several years to deliver specific quantities of nucleic acids directly to the hemolymph of mosquitoes and house flies. This protocol results in minimal injection mortality and allows dose correlated measurements of fecundity.

Here, we present a protocol to produce house fly carboxylesterase proteins in vitro with a baculovirus mediated insect cell expression system and later functionally characterize their roles in metabolizing permethrin, thereby, conferring pyrethroid resistance by conducting cell-based MTT assay and in vitro metabolic studies.

The FlyBar: Administering Alcohol to Flies

Drosophila has emerged as a significant model system for dissecting the cellular and molecular underpinnings of behavioral responses to alcohol. Here we present a protocol for the collection of alcohol sensitivity data in a circadian context that can be easily applied to other experiments and is well-suited for undergraduate research.
