Haploid Diploid Mating

Haploid diploid mating is a reproductive process in which haploid cells or gametes fuse to form a diploid cell, linking two stages of a sexual life cycle. During syngamy, compatible haploid cells combine their nuclei, restoring paired chromosome sets and producing a diploid zygote; subsequent meiosis can reduce chromosome number and generate new haploid cells. This alternation between haploid and diploid states is central to genetic inheritance and variation in fungi, plants, animals, and other eukaryotes. Studying haploid diploid mating helps researchers analyze fertilization, chromosome behavior, recombination, life-cycle regulation, and the genetic basis of traits.

This is a method for generating gynogenetic diploid zebrafish embryos (embryos whose only genetic contribution comes from the mother) by blocking the second meiotic division immediately after fertilization with ultraviolet light-inactivated sperm. EP embryos are not fully homozygous due to recombination during the first meiotic division, however they are homozygous at all loci that have not been separated from their centromere by recombination.

The zebrafish is a powerful model system for developmental biology and human disease research due to their genetic similarity with higher vertebrates. This protocol describes a methodology to create haploid zebrafish embryos that can be utilized for forward screen strategies to identify recessive mutations in genes essential for early embryogenesis.

Sexual Selection Class Simulation ExpandNote: In this activity, you and your classmates will portray a sexually reproducing population consisting of an equal number of male and female animals. You will simulate several different mating scenarios to determine how relative quality, attractiveness, sex, and parental commitment affect mate choice and reproductive success. The goal of this activity is to maximize your progeny. Hypotheses: There are several overall hypotheses for female and male...

Mating and Tetrad Separation of Chlamydomonas reinhardtii for Genetic Analysis

Mating and tetrad separation are required for genetic analysis in Chlamydomonas reinhardtii. Here we demonstrate standard methods for gametogenesis, mating, zygote germination and tetrad dissection. This protocol consists of an easy-to-follow series of steps that will make genetic approaches amenable to scientists who are less familiar with Chlamydomonas.
