Whole Cell Voltage Clamp

Whole-cell voltage clamp is an electrophysiological technique that records ionic currents across the plasma membrane while holding a cell’s membrane potential at a selected value. A glass micropipette forms a high-resistance seal with the membrane, and brief suction or electrical pulses rupture the patch beneath the pipette tip, granting electrical access to the cell interior; the amplifier then injects current to maintain the target voltage and measures the current required. In biology, this method characterizes ion channel activity, membrane conductance, synaptic responses, and action-potential mechanisms, supporting studies of neuronal signaling, muscle physiology, sensory transduction, and drug effects.

Whole-cell recordings from Drosophila melanogaster photoreceptors enable the measurement of spontaneous dark bumps, quantum bumps, macroscopic responses to light, and current-voltage relationships under various conditions. In combination with D. melanogaster genetic manipulation tools, this method enables the study of the ubiquitous inositol-lipid signaling pathway and its target, the TRP channel.

The cut-open Vaseline gap approach is used to obtain low noise recordings of ionic and gating currents from voltage-dependent ion channels expressed in Xenopus oocytes with high resolution of fast channel kinetics. With minor modification, voltage clamp fluorometry can be coupled to the cut-open oocyte protocol.
