Hemophilia A

Hemophilia A is an inherited bleeding disorder in which deficient or impaired clotting factor VIII disrupts normal blood coagulation. Usually caused by mutations in the F8 gene on the X chromosome, it limits formation of the factor VIII–factor IXa complex needed to activate factor X, producing prolonged bleeding after injury and, in severe cases, spontaneous bleeding into joints or muscles. Biology research on hemophilia A supports diagnosis through coagulation testing and genetic analysis, while replacement factor VIII, extended-half-life products, and non-factor therapies help prevent bleeding. Studying its molecular basis also informs gene therapy and improved long-term disease management.

The refined tail vein transection (TVT) bleeding model in anesthetized mice is a sensitive in vivo method for the assessment of hemophilic bleeding. This optimized TVT bleeding model uses blood loss and bleeding time as endpoints, refining other models and avoiding death as an endpoint.
