Hypoxic Chamber Exposure

Hypoxic chamber exposure is a laboratory method that places cells, tissues, animals, or sometimes human participants in an environment with reduced oxygen availability to study responses to hypoxia. By lowering the chamber’s oxygen concentration or oxygen partial pressure under controlled conditions, researchers create a repeatable stimulus that can alter cellular metabolism, blood flow, gene regulation, and stress signaling. In biology, the approach helps clarify how organisms sense and adapt to oxygen limitation, including changes associated with development, exercise, disease, and environmental stress. Controlled exposure also supports comparisons between oxygen levels, exposure durations, and recovery conditions in experimental research.

Hypoxic pulmonary vasoconstriction (HPV) is an important physiological phenomenon by which at alveolar hypoxia lung perfusion is matched to ventilation. The major vascular segment contributing to HPV is the intra-acinar artery. Here, we describe our protocol for the analysis of HPV of murine pulmonary vessels with diameters of 20-100 μm.
