Image J Quantification

ImageJ quantification is the use of ImageJ, an open-source image-analysis platform, to convert biological images into measurable data such as area, length, intensity, and particle number. The process typically involves calibrating pixel dimensions, selecting regions of interest, adjusting image properties or thresholds, and applying measurements or particle-analysis tools to distinguish and quantify structures. Swarming motility is influenced by physical and environmental factors. We describe a two-phase protocol and guidelines to circumvent the challenges commonly associated with swarm assay preparation and data collection. A strategy to quantitatively analyze histological data in the bone marrow is presented. Confocal microscopy of fluorescently labeled cells in tissue sections results in 2-dimensional images, which are automatically analyzed. Co-localization analyses of different cell types are compared to data from simulated images, giving quantitative information about cellular interactions.
