Immune Checkpoint Inhibitors

Immune checkpoint inhibitors are therapies that enhance immune responses by blocking molecular signals that restrain T-cell activity, allowing the immune system to recognize and attack abnormal cells. In cancer biology, antibodies targeting checkpoints such as CTLA-4 or the PD-1/PD-L1 pathway prevent inhibitory receptor-ligand interactions, restoring T-cell activation, proliferation, and cytotoxic function within tumors. These treatments have produced durable responses in several cancers and transformed immunotherapy research, while also revealing important limits, including variable patient responses and immune-related adverse events caused by excessive immune activation. Studying checkpoint signaling supports biomarker development, combination therapies, and safer strategies for regulating antitumor immunity.

Xenopus egg extract is a useful model system to investigate the DNA damage checkpoint. This protocol is for the preparation of Xenopus egg extracts and DNA damage checkpoint inducing reagents. These techniques are adaptable to a variety of DNA damaging approaches in the study of the DNA damage checkpoint signaling.
