Immunogenic Epitope Identification

Immunogenic epitope identification is the process of locating antigenic regions that can trigger recognition and activation by the adaptive immune system. It combines analysis of antigen sequences or structures with experimental testing of peptide–MHC binding, antibody recognition, or immune-cell responses to determine which epitopes are presented and immunogenic under specific biological conditions. In biology, this approach supports vaccine design, diagnostic development, antibody engineering, and studies of host–pathogen interactions. Identifying relevant epitopes can improve the precision of immune assays and help researchers understand how antigen structure, processing, presentation, and immune recognition shape disease and therapeutic responses.

Biochemical assays with recombinant human MHC II molecules can provide rapid, quantitative insights into immunogenic epitope identification, deletion, or design. Here, a peptide-MHC II binding assay scaled to 384-well plates is described. This cost effective format should prove useful in the fields of protein deimmunization and vaccine design and development.

This video showcases an indirect enzyme-linked immunosorbent assay or ELISA sandwich immunocapture assay designed to assess immunogenic glycoprotein levels in vaccines. The assay plate, containing glycoprotein-specific antibodies, is incubated with serially diluted vaccine samples, including reference and test vaccines, followed by immunoassay to quantify glycoprotein content in the test vaccine.

Identification and Characterization of Immunogenic RNA Species in HDM Allergens that Modulate Eosinophilic Lung Inflammation

2020

Environmental allergens such as house dust mites (HDM) often contain microbial substances that activate innate immune responses to regulate allergic inflammation. The protocol presented here demonstrates the identification of dsRNA species in HDM allergens and characterization of their immunogenic activities in modulating eosinophilic lung inflammation.
