Immunoprecipitation Assay

An immunoprecipitation assay is a laboratory method that selectively isolates a target protein, or proteins associated with it, from a complex biological sample. The assay uses a specific antibody to bind the target, after which antibody–protein complexes are captured on protein A or protein G beads, washed to remove nonspecific molecules, and eluted for analysis. Researchers commonly examine the recovered material by Western blotting or mass spectrometry to measure protein abundance or identify interacting partners. In biology, immunoprecipitation supports studies of protein expression, post-translational modifications, signaling pathways, and protein–protein interactions.

In this video, we demonstrate a technique to perform the co-immunoprecipitation of GFP-tagged wild-type and mutated PD-1 proteins using beads coated with anti-GFP antibodies. The bead-bound PD-1-GFP proteins are incubated with SHP2 proteins that interact with PD-1, enabling the identification of specific structural motifs in PD-1 that play a critical role in SHP2 interaction.

Chromatin immunoprecipitation is a powerful technique for the identification of DNA binding sites of Arabidopsis proteins in vivo. This procedure includes chromatin cross-linking and fragmentation, immunoprecipitation with selective antibodies against the protein of interest, and qPCR analysis of bound DNA. We describe a simple ChIP assay optimized for Arabidopsis plants.
