Intestinal Stem Cell Isolation

Intestinal stem cell isolation is a biological method for separating self-renewing stem cells from intestinal tissue so their maintenance, differentiation, and regenerative capacity can be studied in controlled laboratory systems. The process commonly involves collecting intestinal crypts, releasing cells through mechanical and enzymatic dissociation, and enriching or identifying stem cells using features such as LGR5 expression before culture in defined, growth-factor-containing media. Isolated cells can generate intestinal organoids that model epithelial renewal, tissue injury, disease, and responses to candidate therapeutics. This approach supports research in developmental biology, gastrointestinal pathology, and regenerative medicine while enabling analysis of stem cell behavior outside native tissue.

This article describes the technique for isolating pancreatic cancer stem cells (CSCs) from human pancreatic ductal adenocarcinoma (PDAC) tissue. These CSCs are used to study the functional effects of drug treatment on the in vitro self-renewal capacity of CSCs for developing more efficient cancer treatments.
