Intracellular Calcium

Intracellular calcium refers to calcium ions within cells, where tightly regulated changes in concentration act as signals that coordinate essential biological functions. Signals such as hormones, neurotransmitters, or membrane depolarization can open calcium channels in the plasma membrane or release calcium from internal stores, including the endoplasmic reticulum; calcium-binding proteins then translate these changes into cellular responses. Calcium signaling regulates muscle contraction, secretion, gene expression, metabolism, and cell survival. Studying intracellular calcium dynamics helps researchers understand processes ranging from neuronal communication and cardiac function to fertilization, disease mechanisms, and the effects of therapeutic compounds.

We demonstrate how changes in the intracellular free calcium concentration and synaptic efficacy can be simultaneously monitored in a ganglion preparation of Aplysia. We image intracellular calcium using a fluorescent dye, Calcium Orange, and induce and monitor synaptic transmission with sharp (intracellular) electrodes.

Simultaneous Measurement of Intracellular Calcium and Membrane Potential in a Mouse Cerebral Endothelium

2025

The video demonstrates the simultaneous measurement of intracellular calcium levels and changes in membrane potential in endothelial cells of mouse cerebral artery segments. The isolated endothelial tube is loaded with a fluorescent calcium indicator and treated with a drug that induces intracellular calcium release, resulting in a change in membrane potential. The indicator's fluorescence enables the measurement of calcium levels, and a recording electrode is inserted into a cell to measure...

Calcium Imaging in Neurons

2023

Calcium ions play an integral role in neuron function: They act as intracellular signals that can elicit responses such as altered gene expression and neurotransmitter release from synaptic vesicles. Within the cell, calcium concentration is highly dynamic due to the presence of pumps that selectively transport these ions in response to a variety of signals. Calcium imaging takes advantage of intracellular calcium flux to directly visualize calcium signaling in living neurons. This video begins...
