Intracellular Tissue Factor

Intracellular tissue factor is the pool of tissue factor located within cells rather than exposed on the plasma membrane, where it can support coagulation and cell signaling. In resting cells, tissue factor is retained in intracellular compartments and is functionally separated from circulating factor VII; cellular activation, injury, or trafficking can move it to the surface, enabling factor VIIa binding and initiation of the coagulation cascade. Studying this compartment helps clarify how endothelial cells, monocytes, and other tissues regulate thrombin generation, inflammation, angiogenesis, and tissue repair. These insights may improve understanding of thrombosis and the development of targeted therapies.

This article outlines the protocol for quantifying the percentage of Tissue Factor (TF)-positive platelets using whole blood flow cytometry, assessing the protein: (1) intracellularly in resting conditions, and (2) on the cell surface, in both resting and activated conditions. Guidance is also provided for evaluating TF-positive platelets in platelet-rich plasma.

The method described here is used to induce the apoptotic signaling cascade at defined steps in order to dissect the activity of an anti-apoptotic bacterial effector protein. In this protocol a method to measure intracellular protein refolding after heat shock is described. This method can be used to study foldases like molecular chaperones and their co-factors or compounds able to influence their activity. Firefly luciferase activity is used as reporter to measure chaperone refolding activity.

Extracellular Vesicle Tissue Factor Activity Assay

Here we describe an in-house extracellular vesicle tissue factor activity assay. Activity-based assays and antigen-based assays have been used to measure tissue factor in extracellular vesicles from human plasma samples. Activity-based assays have higher sensitivity and specificity than antigen-based assays.
