Ischemic Hypoxia

Ischemic hypoxia is a condition in which reduced blood flow limits oxygen delivery to tissue, combining oxygen deficiency with inadequate nutrient supply and waste removal. When perfusion falls, mitochondrial respiration and ATP production decline, while oxygen-sensitive pathways stabilize hypoxia-inducible factors that alter gene expression, metabolism, and vascular signaling. These responses can support short-term adaptation by promoting anaerobic metabolism and angiogenesis, but prolonged ischemic hypoxia causes cellular dysfunction, inflammation, and tissue injury. Studying this process is essential for understanding stroke, myocardial infarction, peripheral vascular disease, wound repair, and how tumors adapt to poorly perfused environments.

Modeling Hypoxia/Reoxygenation Injury in Proximal Tubular Epithelial Cells

Here, we describe an in vitro protocol to model hypoxia/reoxygenation damage in the highly metabolic proximal tubular epithelial cells. This model is designed to induce metabolic stress-induced damage, which can be measured through the expression of proximal tubule damage markers and assessment of mitochondrial respiratory function.

Thromboembolic stroke models are vital tools for optimizing the recanalization therapy. Here we report a murine thrombotic stroke model based on transient cerebral hypoxic-ischemic (tHI) insult, which triggers thrombosis and infarction, and responds favorably to tissue plasminogen activator (tPA)-mediated fibrinolysis in a therapeutic window similar to those in stroke patients.
