Kidney Capsule

The kidney capsule is a thin, tough layer of connective tissue that surrounds each kidney and helps protect the organ while maintaining its shape. Composed mainly of collagen-rich fibers, it forms a close-fitting outer covering beneath the surrounding fat and connective tissues, resisting mechanical stress and limiting the spread of inflammation or infection into nearby structures. In biology, studying the kidney capsule helps explain renal anatomy, tissue organization, and the physical protection of nephrons and blood vessels. Changes in the capsule can also provide clues about kidney injury, inflammation, scarring, or disease-related enlargement.

In this video, we describe the procedure for kidney extraction from the retroperitoneal space of the mouse. The extracted kidney can further be microdissected to obtain an intact kidney without a renal capsule and the surrounding fat layer.

Our protocol was developed to cleanly and easily deliver islets or cells under the kidney capsule of mice. Cells are concentrated into pellets in the final tubing used for transplanting the cells under the kidney capsule. The ease of this technique reduces stress to the cells and the mouse.
