Antibody Incubation

Antibody incubation is a laboratory step in which antibodies are allowed to bind specific target molecules, enabling their detection or measurement in biological samples. During incubation, primary antibodies recognize and attach to antigens through complementary molecular interactions, while controlled time, temperature, buffer composition, and blocking conditions promote specific binding and limit nonspecific background; secondary antibodies may then bind the primary antibody to amplify the signal. This technique supports immunofluorescence, immunohistochemistry, Western blotting, and other immunoassays. Careful optimization of incubation conditions improves signal clarity, assay sensitivity, and reproducibility in studies of protein localization, expression, and cellular function.

2022

Laboratory soil warming experiments usually employ two or more constant temperatures in multiple chambers. By presenting a sophisticated environmental chamber, we provide an accurate temperature control method to imitate the magnitude and amplitude of in situ soil temperature and improve the experimental design of soil incubation studies.

Scalable High Throughput Selection From Phage-displayed Synthetic Antibody Libraries

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.

A General Method for Evaluating Incubation of Sucrose Craving in Rats

Responding for food or drug-paired cues increases over the course of a period of abstinence and this may relate to an increased susceptibility to relapse behaviors. Here we detail a procedure for evaluating this "incubation of craving" in rats that have self-administered sucrose.

A Technique for the Prolonged Incubation of Neuronal Tissue of the Retina

2025

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.
