Dual Cell Recording

Dual cell recording is an electrophysiological method that measures the activity of two cells simultaneously, allowing researchers to examine communication between individual neurons or other excitable cells. Typically, researchers establish whole-cell patch-clamp recordings with two electrodes, stimulate one cell by injecting current or eliciting action potentials, and monitor voltage or synaptic currents in the other; the timing and properties of the response reveal functional connectivity. In biology, this approach helps characterize synaptic transmission, connection strength, excitatory and inhibitory interactions, and the cellular mechanisms underlying neural circuits. It is especially valuable for linking individual cell properties to network function and circuit development.

Dual Electrophysiological Recordings of Synaptically-evoked Astroglial and Neuronal Responses in Acute Hippocampal Slices

The preparation of acute brain slices from isolated hippocampi, as well as the simultaneous electrophysiological recordings of astrocytes and neurons in stratum radiatum during stimulation of schaffer collaterals is described. The pharmacological isolation of astroglial potassium and glutamate transporter currents is demonstrated.

Activity in neuronal systems often requires synchronous action potential discharges from neurons within a specific population. For example, pulses of gonadotropin-releasing hormone (GnRH) likely require coordinated activity between GnRH neurons. We present our methodological approach for reliably obtaining simultaneous electrophysiological recordings from the diffusely distributed GnRH neurons.
