Lung Microvasculature

Lung microvasculature is the dense network of small blood vessels that supports pulmonary circulation and enables efficient exchange between blood and the air-filled alveoli. Its thin capillary walls bring deoxygenated blood into close contact with alveolar epithelium, allowing oxygen and carbon dioxide to diffuse across the alveolar-capillary barrier while endothelial cells regulate fluid movement and vascular tone. This specialized structure is essential for maintaining blood oxygenation and matching ventilation with perfusion. Studying lung microvasculature helps researchers understand pulmonary edema, inflammation, thrombosis, and vascular remodeling, while informing research on respiratory disease, drug delivery, tissue engineering, and regenerative medicine.

Two-Photon Microscopy for In Vivo Imaging of Mouse Brain Microvasculature

2025

Source: Nishimura, C. et al. Quantification of Cerebral Vascular Architecture using Two-photon Microscopy in a Mouse Model of HIV-induced Neuroinflammation. J. Vis. Exp. (2016). This video demonstrates the in vivo visualization of cortical capillaries in the mouse brain using two-photon microscopy. It outlines the steps for injecting a fluorescent dye into an anesthetized mouse with a thin-skull cortical window, preparing the setup for imaging, and acquiring detailed images of brain...

Spatial Temporal Analysis of Fieldwise Flow in Microvasculature

To quantify microvascular flow from high speed capillary flow image sequences, we developed STAFF (Spatial Temporal Analysis of Fieldwise Flow) software. Across the full image field and over time, STAFF evaluates flow velocities and generates a sequence of color-coded spatial maps for visualization and tabular output for quantitative analyses.

DCE-MRI of Orthotopic Pancreatic Tumor Xenografts: A Method to Assess Microvasculature in a Target Tumor Tissue

2023

This video describes the technique of DCE-MRI for orthotropic pancreatic tumor xenografts in mice. DCE-MRI is a non-invasive method to analyze microvasculature in a target tissue, and useful to assess vascular response in a tumor following a novel therapy. This method will assist investigators to apply DCE-MRI for orthotopic gastrointestinal cancer mouse models.

This paper describes a striaghforward and efficient method of intubating mice for pulmonary function measurements or pulmonary instillation, that allows the mice to recover and be studied at later times. The procedure involves an inexpensive fiberoptic light source that directly illuminates the trachea.

Lung Capacity

2019

The air in the lungs is measured in volumes and capacities. Lung volume measures reflect the amount of air taken in, released, or left over after a lung function, like a single inhalation. Lung capacity measures are sums of two or more lung volume measures. There are four defined lung volume measures: tidal volume, inspiratory reserve volume, expiratory reserve volume, and residual volume. Tidal volume is the amount of air inhaled and exhaled in a normal breath. Inspiratory...
