Mouse Embryonic Stem Cells

Mouse embryonic stem cells are pluripotent cells derived from the inner cell mass of early mouse blastocysts, providing a tractable system for studying how cells maintain identity and generate diverse tissues. In culture, signaling conditions such as leukemia inhibitory factor support self-renewal, while changes in growth factors and extracellular cues can initiate differentiation into specialized cell types. Researchers use these cells to investigate early development, model disease, test gene function through targeted genetic modification, and evaluate methods for directed differentiation. Their reproducibility and experimental flexibility have made them a foundational tool in mammalian biology and a platform for advancing stem-cell-based research.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

We developed a new protocol to improve efficiency of in vitro differentiation of mouse embryonic stem cells into motor neurons. The differentiated ES cells acquired motor neurons features as evidenced by expression of neuronal and motor neuron markers using immunohistochemical techniques.

Embryonic stem (ES) cells are undifferentiated pluripotent cells, meaning they can produce any cell type in the body. This gives them tremendous potential in science and medicine since they can generate specific cell types for use in research or to replace body cells lost due to damage or disease. Origin ES cells are present in the inner cell mass of an embryo at the blastocyst stage, which occurs at about 3–5 days after fertilization in humans before the embryo is implanted in the uterus.
