Multicellular Magnetotactic Bacteria

Multicellular magnetotactic bacteria are organized groups of genetically identical bacterial cells that move as a coordinated unit along Earth’s magnetic field, making them a distinctive form of microbial life. Each cell produces membrane-bound magnetosomes containing magnetic minerals, and the aligned magnetosomes collectively orient the consortium through magnetotaxis toward chemically favorable zones, often near oxygen gradients in aquatic sediments. Their coordinated structure and directed movement provide insight into cellular cooperation, biomineralization, and microbial ecology. Studying these bacteria also informs research on biogeochemical cycling, the evolution of multicellularity, and possible biosignatures in Earth’s extreme environments or extraterrestrial settings.

Collection, Isolation and Enrichment of Naturally Occurring Magnetotactic Bacteria from the Environment

We demonstrate a method to collect magnetotactic bacteria (MTB) that can be applied to natural waters. MTB can be isolated and enriched from sediment samples using a relatively simple setup that takes advantage of the bacteria's natural magnetism. Isolated MTB can then be examined in detail using both light and electron microscopy.

We present a procedure for growing several strains of Magnetospirillum in two different types of growth media. Magnetospirillum gryphiswaldense strain MSR-1 is grown in both liquid and O2 concentration gradient semi-solid media while M. magneticum strain AMB-1 and M. magnetotacticum strain MS-1 are grown in liquid medium.

Recording Multicellular Behavior in Myxococcus xanthus Biofilms using Time-lapse Microcinematography

To study Myxococcus xanthus swarm behavior, we have designed a time-lapse microcinematography protocol that can be modified for different assays. It employs standard growth conditions adapted for microscopy, and yields reproducible results by the use of inexpensive, reusable silicone gaskets. We have used this method to quantify multicellular chemotaxis.
