Murine Embryonic Gut

Murine embryonic gut refers to the developing gastrointestinal tract of mouse embryos, a tractable model for studying organ formation and intestinal development. During embryogenesis, coordinated signaling between endoderm-derived epithelium and surrounding mesenchyme drives regional patterning, epithelial differentiation, tissue folding, and elongation. Researchers examine these processes through embryonic tissue culture, organ culture, imaging, and molecular analysis to determine how cells establish intestinal architecture. This model helps clarify fundamental principles of vertebrate development and provides a foundation for investigating congenital gastrointestinal disorders, epithelial biology, and strategies for tissue engineering and regenerative medicine.

Isolation of Murine Embryonic Hemogenic Endothelial Cells

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.

We present a protocol to dissect and culture embryonic day 15 (E15) murine metatarsal bones. This highly physiological ex vivo model of endochondral ossification provides conditions closer to the in vivo situation than cells in monolayer or 3D culture and is a vital tool for investigating bone growth and development.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
