Neural Stem Cells

Neural stem cells are self-renewing, multipotent cells that generate the major cell types of the nervous system, making them central to nervous system development, maintenance, and repair research. They divide asymmetrically or symmetrically and respond to signals within specialized niches, balancing continued self-renewal with differentiation into neurons, astrocytes, and oligodendrocytes. In biology, researchers study neural stem cells to understand brain development, neural regeneration, and disease mechanisms, while cultured cells and stem-cell-derived models support investigations of neurodegenerative disorders and potential cell-based therapies. Their behavior also provides insight into how local environments regulate cell fate and tissue organization.

Here is a protocol to grow pluripotent stem cells (PSC) and neural stem cells (NSC) in an enclosed cell culture system that permits maximum sterility and reproducibility, replacing the traditional biosafety cabinet and incubator. This equipment meets clinical good manufacturing practice (cGMP) and clinical good lab practice (cGLP) guidelines.

Neural crest (NC) cells derived from human pluripotent stem cells (hPSC) have great potential for modeling human development and disease and for cell replacement therapies. Here, a feeder-free adaptation of the currently widely used in vitro differentiation protocol for the derivation of NC cells from hPSCs is presented.
