Optical Density Measurement

Optical density measurement is a spectrophotometric method that estimates the amount of light absorbed or scattered by a sample, providing an indirect measure of its concentration. In biology, a spectrophotometer directs light through a sample at a selected wavelength and records the reduction in transmitted light as absorbance or optical density; under suitable conditions, this signal relates to cell number or biomolecule abundance. Researchers commonly monitor microbial growth by measuring optical density near 600 nm, construct growth curves, and standardize cultures for experiments. The method is rapid and non-destructive, although turbidity, path length, and sample concentration can affect accuracy.
