Osteogenic Differentiation

Osteogenic differentiation is the biological process by which progenitor or stem cells acquire the specialized characteristics of osteoblasts, the cells responsible for forming bone. In response to biochemical signals and culture conditions, pathways such as bone morphogenetic protein and Wnt signaling activate transcription factors including RUNX2 and osterix, promoting extracellular matrix production followed by mineralization. In biology and regenerative research, this process helps evaluate bone-forming potential, optimize scaffolds and cell-based therapies, and model skeletal development or disease. It also supports drug screening and the study of how mechanical, chemical, and cellular cues regulate bone tissue formation.

Quantitative measurement of bone progenitor function in fracture healing requires high resolution serial imaging technology. Here, protocols are provided for using intravital microscopy and osteo-lineage tracking to sequentially image and quantify the migration, proliferation and differentiation of endogenous osteogenic stem/progenitor cells in the process of repairing bone fracture.

Polyelectrolyte Complex for Heparin Binding Domain Osteogenic Growth Factor Delivery

Self-assembled polyelectrolyte complexes (PEC) fabricated from heparin and protamine were deposited on alginate beads to entrap and regulate the release of osteogenic growth factors. This delivery strategy enables a 20-fold reduction of BMP-2 dose in spinal fusion applications. This article illustrates the benefits and fabrication of PECs.

The video demonstrates the differentiation of immortalized multipotent otic progenitors (iMOPs) into spiral ganglion neurons (SGNs) and immunofluorescence-based confirmation of the differentiation. The iMOPs are plated onto culture substrate-coated coverslips in a neuronal differentiation medium for differentiation into SGNs. The differentiated cells are labeled with antibodies specific for neuronal differentiation markers and analyzed under a microscope to confirm differentiation.
