Respirometric Assessment

Respirometric assessment is the measurement of respiration by tracking oxygen consumption, carbon dioxide production, or related changes in gas composition, providing a quantitative view of biological energy metabolism. In a respirometer, a sealed or flow-through chamber monitors gas exchange with sensors while temperature, substrate availability, and other conditions are controlled. The resulting respiration rate reflects cellular or organismal metabolic activity and can reveal responses to environmental stress, nutrients, toxins, or changing physiological demands. In biology, respirometric assessment supports studies of microbial growth, mitochondrial function, soil activity, aquatic organisms, and ecosystem processes, while enabling comparisons of metabolic performance across samples and experimental conditions.

Here, we describe a detailed method for mitochondria isolation from mouse skeletal muscle and the subsequent analysis of respiration by Oxygen Consumption Rate (OCR) using microplate-based respirometric assays. This pipeline can be applied to study the effects of multiple environmental or genetic interventions on mitochondrial metabolism.
