Paracrine Signaling

Paracrine signaling is a form of cell communication in which a cell releases chemical messengers that act on nearby cells, coordinating local biological activities within tissues. Signaling molecules such as growth factors, cytokines, and neurotransmitters diffuse through the extracellular space and bind receptors on neighboring target cells, triggering intracellular pathways that alter gene expression, metabolism, proliferation, or movement. This short-range communication supports embryonic development, immune responses, tissue repair, and maintenance of tissue organization. Studying paracrine signaling helps explain how cells interact in healthy tissues and how disrupted local signals contribute to diseases including cancer, inflammation, and fibrosis.

In this video, we develop a proximal co-culture model of ovarian cancer cells and primary human mesothelial cells. This physiologically relevant model allows the study of paracrine signaling, where the localized concentrations of the secreted factors are maintained while preventing direct cellular contact.

An efficient method to gain insights into visualizing the paracrine-derived ROS induction of endothelial Ca2+ signaling is described. This method takes advantage of measuring paracrine derived ROS triggered Ca2+ mobilization in vascular endothelial cells in a co-culture model.
