Pbs Wash

PBS wash is a laboratory technique that uses phosphate-buffered saline to rinse cells, tissues, or assay surfaces while maintaining conditions compatible with biological samples. Its phosphate buffer helps keep pH stable, while sodium chloride and related salts provide an approximately isotonic solution that removes residual media, unbound reagents, and debris without causing substantial osmotic stress. In cell culture, immunostaining, enzyme-linked immunosorbent assays, and protein workflows, researchers typically add PBS, gently mix or rock the sample, and remove the liquid before repeating as needed. Proper washing improves signal-to-noise, reduces background, and supports reproducible experimental results.

After filtration, the precipitate is washed to remove coprecipitated impurities and any remaining mother liquor. We describe a straightforward method for the isolation of washed platelets from human blood followed by agonist-induced platelet aggregation measurements by turbidimetry. As an example we apply this method for studying the aggregation response of human platelets to collagen after a pre-incubation with the Pannexin1 channel inhibitor Brilliant Blue FCF.
